Antitumor, toxicity and target gene expression evaluation of MiR-204-5p mimic application on melanoma b16-bearing mice

Objective. To evaluate anti-tumor, toxic effect of miR-204-5p mimic applicaton on melanoma B-16-bearing mice followed by target gene expression estimation in tumor and distant organs. Material Methods. C57Bl/6 were used. The animals the experimental group intraperitoneally injected with a 5 nM miRNA simulator (mimic) 8th, 10th, 12th days after B-16 cell transplantation. Based results bioinformatic analysis, genes BCL2 SIRT1 levels determined quantitative real-time PCR. was estimated evaluation body weight, mass internal organs, motor activity. Results. On 13-14th experiment, activity control groups decreased signifcantly compared to treated miR-204-5p. Target showed increased lungs kidneys (p˂0.05). Tumor tended decrease mimic. Conclusion. Modulation level microRNA led changes animals, BCL2in kidneys. MiR-204-5p application did not have treated. Further studies are necessary clarify implication proliferation regulation as well it’s biodistibution tissue.
